Research on the inhibitory effects of galangin on the metastasis of breast cancer and related mechanisms

Objective To explore the effects of galangin on the migration and invasion of MDA-MB-231 breast cancer cells and its mechanisms. Methods MTT assay was adopted to observe the effects of galangin at different concentrations on the proliferation of MDA-MB-231 breast cancer cells. The wound-healing and Transwell assays were conducted to investigate the effects of galangin on the horizontal and vertical migration of MDA-MB-231 cells. The Transwell chamber system was used to investigate the effects of galangin on the invasion of MDA-MB-231 cells. Western blotting was adopted to examine the effects of galangin on the levels of PI3K, Akt, MMP-2, and MMP-9 in MDA-MB-231 cells. Meanwhile, immunofluorescence assay was adopted to explore effects of galangin on the levels of PI3K, Akt, MMP-2, and MMP-9 in MDA-MB-231 cells. The influences of galangin on nuclear transcription factor (NF-κb) p65 phosphorylation and nuclear translocation were examined by Western blotting and immunofluorescence assay. Furthermore, a xenograft tumor model of nude mouse was established using MDA-MB-231 breast cancer cell line. Meanwhile, 200 μg/kg galangin was intragastrically (ig) administrated for 15 days and the size and weight of each tumor were measured to calculate the inhibitory rate of tumor. The amounts of PI3K, AKT, MMP-2, MMP-9 and p-NF-κb p65 in tumor tissues were determined by Western blotting. Results The proliferation of MDA-MB-231 cells could be remarkably inhibited by galangin at 50, 100 and 200 μmol/L in vitro. Exposure to galangin could suppress the quantities of PI3K, Akt, MMP-2 and MMP-9 proteins in a dose-dependent manner and relieve the phosphorylation and nuclear translocation of NF-κb p65. The growth of breast cancer was blocked in nude mice by 200 μmol/L galangin, with an inhabitation rate of 43.66%. Galangin could decrease the levels of PI3K, Akt, MMP-2, and MMP-9 and p-NF-κb p65 in tumor tissues. Conclusions Galangin can significantly inhibit the growth of MDA-MB-231 cells in vivo, and block the proliferation, migration and invasion of the cells in vitro, suppress the levels of related proteins like PI3K, Akt, MMP-2, and MMP-9 and block the activation of transcription factor NF-κb p65 through inhibiting its phosphorylation and nuclear translocation.
